Biological Response: Biliary Tract Health & Bitter Botanicals
The biliary tract is comprised of the liver, gallbladder, and a network of bile ducts whose synergistic activity regulates the synthesis, storage, and movement of bile. Importantly, the biliary tract functions as the singular route by which bile is transported to the duodenum, the first segment of the small intestine. This labyrinthine passage is often called the biliary tree because it begins deep within the liver as many small branches (canaliculi) that merge with progressively larger hepatic and gallbladder ducts
to form the trunk-like common bile duct. This centralized duct is joined by the pancreatic duct further along, where sphincters surrounding them control the flow of bile and pancreatic ‘juices’ (enzymes, bicarbonate) into the duodenum.
Bile, a dark green to yellowish-brown fluid, is composed of water (∽98%), electrolytes, cholesterol, phospholipids, conjugated bilirubin, and bile salts. Most of its components are extracted from circulating blood by the liver. Bile acids are synthesized by hepatocytes (liver cells) through the metabolism of cholesterol. Their subsequent conjugation with water-soluble amino acids creates bile salts Hydrangea arborescens that are released into intrahepatic bile ducts by the bile salt export pump (BSEP), a crucial biliary transport protein.

Electrolytes are actively transported into bile ducts; water enters them through osmosis. Bilirubin, a toxic byproduct of hemoglobin metabolism, is carried there from hepatocytes by MRP2, another transport protein.
Once formed, bile travels through the hepatic duct system to the gallbladder, where it is stored. During a meal, smooth muscle within the gallbladder wall contracts, propelling bile into the common bile duct and duodenum where it facilitates the digestion of dietary fats, absorption of fat-soluble vitamins (A, D, E, K), and excretion of bilirubin. Most of the bile salts are returned to the liver by enterohepatic circulation for repeated reuse.

Phytochemistry
Most of the botanicals in Core Hydrangea Blend support hepatobiliary health through cholagogic and choleretic activity. Cholagogues are herbs whose bitter compounds sustain liver and gallbladder function by encouraging the flow of bile into the duodenum. Long-chain fatty acids, amino acids, flavonoids, saponins, and terpenoids trigger the release of cholecystokinin (CCK), a peptide hormone synthesized by duodenal mucosa that promotes gallbladder contraction and biliary sphincter relaxation.
Many of these plants are also recognized as choleretics whose compounds increase bile formation through various mechanisms. The hormone CCK stimulates secretion of bile salts into hepatic canaliculi via signaling with the transport protein BSEP. Certain flavonoids increase the expression of biliary transport proteins and engage with a signaling pathway known as cyclic AMP that boosts bile acid production by hepatocytes. Flavonoids, terpenes, and vitamin C protect biliary tract cells from oxidative stress primarily through free radical scavenging. They also encourage the natural production of antioxidant enzymes such as superoxide dismutase; trace minerals like copper, manganese, and zinc act as cofactors that activate them. Quercetin, a flavonoid found in hydrangea and borage, and chlorogenic acid, a caffeic acid ester plentiful in Jerusalem artichoke, raise cellular levels of glutathione. An antioxidant vital to hepatic and renal mitochondrial function, glutathione scavenges free radicals and recycles vitamins C and E. Flavonoids, terpenes, alkaloids, and saponins normalize bile flow through relaxation of smooth muscle within bile ducts, gallbladder wall, and related sphincters.

Total body water is distributed between intracellular fluid (ICF) and extracellular fluid (ECF) compartments. Two-thirds of this water is contained in ICF and the remainder in ECF. These compartments are not literal anatomical chambers; they
merely illustrate the division of water and its solutes (dissolved substances) throughout the body. ECF is subdivided further into the interstitial fluid that surrounds cells and intravascular fluid, the serum held by blood vessels. In healthy individuals, the dynamic equilibrium between ICF and ECF is established by homeostatic control mechanisms including feedback loops, electrolyte-induced pressure gradients, cardiovascular prompts, neuroendocrine signaling and, above all, the production of urine by the kidney.
The kidney participates in systemic homeostasis by regulating blood pressure, serum pH, excretion of metabolic waste, electrolyte concentrations, and extracellular fluid volume—all of which begins with its filtration of blood. Nephrons, the functional units of the kidney, contain tufts of capillaries called glomeruli that filter blood passively, releasing small molecules like glucose, water, and amino acids into renal tubules while blocking passage of larger proteins and cells. This glomerular filtrate is processed further by a system of tubules and collecting ducts whose epithelial lining houses various transport proteins. Through complex hormonal signaling these proteins move water, electrolytes (e.g. sodium, potassium, calcium), nutrients, and metabolites back and forth between filtrate and blood. Useful substances like water, electrolytes, amino acids, and glucose are reabsorbed before the remaining fluid—composed mostly of water, metabolic waste (e.g. urea, creatinine, uric acid), and pigments—is released into the bladder as urine.

Phytochemistry
Many of the botanicals in Core Hydrangea Blend are rich in compounds that support fluid and electrolyte balance at the nephron level. Flavonoids, triterpenes, and tannins modulate urine composition, volume, and flow by promoting the natural excretion of water and electrolytes by tubules and collecting ducts. Mucilage and inulin, polysaccharides plentiful in borage, Jerusalem artichoke, and blessed thistle, help maintain urine volume and flow through an osmotic attraction of water.
Flavonoids like quercetin reduce the expression of epithelial sodium channel, a transport protein that moves sodium ions across distal tubule membranes. Flavonoids activate sodium-potassium-chloride cotransporter 2 (NKCC2), a transport protein that shuttles sodium, potassium, and chloride ions between tubule fluid and blood. Through its selective mediation of sodium reabsorption, NKCC2 regulates electrolyte concentrations and extracellular fluid volume. Flavonoids calm the sodium-potassium pump, a cell membrane enzyme that balances fluid and electrolytes within renal tubules.

Certain dietary substances including calcium and oxalate tend to crystallize and form insoluble salts in the urine. The presence of uric acid (a byproduct of purine metabolism), coupled with low levels of urinary citrate, quickens this process. Mucilage and inulin, which are soluble fiber, bind with calcium and oxalate in the small intestine, shifting their elimination from kidney to colon. Calcium, naturally abundant in most of the botanicals in Core Hydrangea Blend, complexes with oxalates in the small intestine as well.
Magnesium, a key electrolyte prevalent in clubmoss, borage, Jerusalem artichoke, yellow dock, and blessed thistle, inhibits crystallization by forming a more soluble salt with oxalate and increasing water solubility of calcium. Potassium, another vital electrolyte, reduces urinary calcium levels by increasing its tubular reabsorption.
Flavonoids, triterpenes, saponins, and tannins inhibit the deposition and growth of calcium oxalate crystals by restoring normal urinary citrate and magnesium levels. Flavonoids increase urinary excretion of oxalate and uric acid; by maintaining an acidic pH, they support potassium and magnesium balance. Alkaloids, coumarins, terpenoids, and flavonoids encourage smooth muscle relaxation, enabling insoluble particles to be flushed from the kidney and bladder more easily.
Flavonoids, tocopherols, and vitamin C are potent free radical scavengers that protect renal tissue on multiple levels. Flavonoids and isothiocyanates enhance the activity of phase II antioxidant enzymes that protect cellular components from oxidative stress. Flavonoids like quercetin, apigenin, and rutin encourage the natural production of cellular antioxidants such as superoxide dismutase and glutathione that shield cell membranes from lipid peroxidation. Rutin, a flavonoid abundant in radish seed and hydrangea root, strengthens glomerular capillaries via molecular signaling with glutathione pathways.
